Certain tumour viruses (including both DNA and RNA viruses) carry genes, which confer on them the ability to convert host cells to a tumorigenic state. These genes are called oncogenes (Table 45.1). Such DNA and RNA viruses differ in some important characteristics which will be briefly described […] (b) RNA viruses. RNA tumour viruses may be transformation competent (able to transform the infected cells) or transformation-incompetent (able to replicate, but not transform the infected cells) […] Horizontal and vertical transfer of genetic information from retroviruses; horizontal transfer involves infection of new host, while vertical transfer involves transfer from one generation to the next generation, after integration of viral genome in the germ line of the host.

Gene Therapy Defined

Gene therapy refers to the replacement of faulty genes, or the addition of new genes as a means to cure disease or improve the ability to fight disease. Genome editing is one aspect of gene therapy. Established approaches to gene therapy have been based on the results of extensive prior laboratory research on individual cells and on nonhuman organisms, establishing the means to add, delete, or modify genes in living organisms. Key advances include the development of techniques for generating molecular tools for cutting the DNA of genomes in specific places to allow targeted alterations in the DNA sequence. Over recent years, several such methods have been introduced and used effectively in clinical applications.

Methods for Producing Recombinant Coronavirus

Abstract

A helper cell for producing an infectious, replication defective, coronavirus (or more generally nidovirus) particle cell comprises (a) a nidovirus permissive cell; (b) a nidovirus replicon RNA comprising the nidovirus packaging signal and a heterologous RNA sequence, wherein the replicon RNA further lacks a sequence encoding at least one nidovirus structural protein; and (c) at least one separate helper RNA encoding the at least one structural protein absent from the replicon RNA, the helper RNA(s) lacking the nidovirus packaging signal. The combined expression of the replicon RNA and the helper RNA in the nidovirus permissive cell produces an assembled nidovirus particle which comprises the heterologous RNA sequence, is able to infect a cell, and is unable to complete viral replication in the absence of the helper RNA due to the absence of the structural protein coding sequence in the packaged replicon. Compositions for use in making such helper cells, along with viral particles produced from such cells, compositions of such viral particles, and methods of making and using such viral particles, are also disclosed.
